Olink Plasma Proteomics Data

Relative plasma protein abundance was measured using the Olink Explore 1536 platform in multiple batches. To normalize across batches, we used a common set of samples included in each batch, and used these common samples to calculate batch normalization offsets. The offsets learned from the batch controls were then applied across all assays in each batch.

Our Olink datasets include the original normalized protein expression (NPX) and limit of detection (LOD) values reported by Olink, as well as these offset values and batch corrected NPX and LOD.

The plasma proteomics dataset contains some sample-level metadata to enable connection between these results and samples used in other modalities. The sample.sampleKitGuid identifier is used across many data modalities:

Sample Identifiers
sample.sampleKitGuid: A Globally Unique Identifier (GUID) for the Sample Kit, representing all material collected at a visit
specimen.specimenGuid: A GUID for the Specimen, representing the specific aliquot of plasma that was measured

Olink Data
olink.batch_id: A unique ID specifying each batch of samples submitted together
olink.plate_id: A unique ID for the specific plate the samples were aliquoted into
olink.panel: The name of the panel of Olink assays (Cardiometabolic, Inflammation, Oncology, or Neurology)
olink.assay: Protein name/symbol for the specific Olink assay (e.g. TNFRSF1B, IL7R)
olink.assay_id: A unique ID for the specific Olink assay (e.g. OID21145, OID21136)
olink.uniprot_id: The Uniprot ID for the protein assayed (e.g. P20333, P16871)
olink.norm_offset: The offset value used to normalize between batches
olink.NPX_raw: The normalized protein expression (NPX) value reported by Olink
olink.NPX_norm: The batch-normalized NPX value after adjusting with the olink.norm_offset value
olink.LOD_raw: The limit of detection (LOD) value reported by Olink
olink.LOD_norm: The batch-normalized LOD value after adjusting with the olink.norm_offset value

Subject Identifiers and Metadata (prefix: cohort or subject)
cohort.cohortGuid: A GUID for the Cohort the subject enrolled in for our study
subject.subjectGuid: A GUID for the Subject
subject.biologicalSex: The biological sex of the Subject
subject.birthYear: The Birth Year of the Subject
subject.ageGroup: The Age Group that the subject belongs to (Pediatric, Young Adult, or Older Adult)
subject.race: The self-reported Race of the Subject
subject.ethnicity: The self-reported Ethnicity of the subject
subject.cmv: The CMV status of the subject (Positive or Negative)

Sample Metadata (prefix: sample)
sample.visitName: The name of the study visit (i.e. time point)
sample.subjectAgeYears: The age of the Subject in years at the time of sample collection
